Reboxetine – a ‘NaRI’ Clarify desipramine

چکیده

Where is its place in therapy? ! There is no convincing evidence to suggest an overall advantage in efficacy, relapse prevention or speed of onset with reboxetine over existing antidepressants. ! It may be useful in SSRI resistant depression. More evidence is needed to support its use in indications such as anxiety, OCD and panic disorder. ! Noradrenergic deficiency differs to serotonergic deficiency and is associated with impaired attention, problems concentrating, fatigue and memory deficits. These symptoms may respond best to treatment with a selective noradrenergic agent, such as reboxetine. ! Like the SSRIs, reboxetine offers advantages over the tricyclics and the MAOIs in terms of safety. ! Unlike the SSRIs & TCAs it does not significantly interact with cytochrome P450 enzymes. ! Reboxetine is associated with “pseudo-anticholinergic” symptoms (not due to direct action on cholinergic receptors). This may preclude its use in certain patients.

Effects of reboxetine on sympathetic neuroeffector transmission in rabbit carotid artery.

The effect of reboxetine on sympathetic neuroeffector transmission in rabbit isolated carotid artery was examined. Reboxetine (10-8-3 x 10-6 M) and cocaine (10-6-3 x 10-5 M), but not desipramine (10-8-3 x 10-7 M), increased contractions evoked by electrical field stimulation. At higher concentrations, reboxetine (10-4 M), cocaine (3 x 10-4 M), and desipramine (3 x 10-7-10-5 M) inhibited the neu...

Therapeutic effects of escitalopram and reboxetine in seasonal affective disorder: a pooled analysis.

The monoaminergic neurotransmitters serotonin and noradrenaline have both been implicated in the pathogenesis of seasonal affective disorder (SAD). However, the differential therapeutic value of selective serotonin reuptake inhibitors (SSRI) and selective noradrenaline reuptake inhibitors (NARI) in SAD has not been assessed until now. This study compares data from two open-label trials with sim...

Potentiation of the antidepressant-like effect of desipramine or reboxetine by metyrapone in the forced swimming test in rats.

The obtained results revealed that joint administration of desipramine or reboxetine and metyrapone (a glucocorticoid synthesis inhibitor) had more potent antidepressant-like activity in the forced swimming test (FST) in rats compared to treatment with either drug alone. WAY100636 (a 5-HT(1A) antagonist), and prazosin (an alpha(1)-adrenergic antagonist), used in doses ineffective in the FST, in...

[Reboxetine in the treatment of bulimia nervosa].

There is a substancial body of literature demonstrating the efficacy of selective serotonin reuptake inhibitors antidepressants (SSRI) in reducing binge eating and vomiting frequency in Bulimia Nervosa. Good therapeutic response to noradrenergic agents, like desipramine and reboxetine, though not frequently reported in literature, has already been demonstrated. This case report describes the tr...

Reboxetine: functional inhibition of monoamine transporters and nicotinic acetylcholine receptors.

The present study determined whether repeated administration of the antidepressant and selective norepinephrine (NE) uptake inhibitor reboxetine resulted in an adaptive modification of the function of the NE transporters (NETs), serotonin (5-HT) transporters, or dopamine (DA) transporters. Because antidepressants may be effective tobacco smoking cessation agents and because antidepressants have...
